I think the thesis you're getting at here could be that Greyhound is saying that women have to be efficient and careful in everything that they do, so if they're going to travel alone, they should do it with Greyhound. Also, I feel like the ad is specifically catering to white women since that's the woman they show, and this is during a time of segregation on buses, so it could be kind of contributing to the idea of segregation too.

Greyhound reinforces the idea that their service is friendly to women by stating that women are the largest demographic of its users.
